What you can expect

Our research focuses on congenital vascular malformations, rare diseases primarily caused by somatic mutations, with limited treatment options. Arteriovenous malformations (AVM) have the most serious medical implications within vascular malformations, with challenging treatment prospects. AVMs have been shown to exhibit mosaic-activating variants in oncogenes along the RAS/MAPK signalling pathway, but show a significant phenotypic variability. Hence, our next step involves expanding knowledge on causal somatic mutations in extra-cranial AVM. We aim to investigate expression patterns at the RNA level and their spatial distribution in diseased tissue, connecting patient phenotypes to cellular phenotypes and genotypes in a translational context.

Your Profile

We are looking for a PhD candidate to establish single cell sequencing in affected endothelial cells (EC) and characterize potential new targets identified by single cell sequencing. In parallel spatial transcriptomics in formalin-fixed paraffin-embedded (FFPE) tissue slides of the same patient tissues to obtain information on the spatial distribution of RNA expression are planned. Ultimately, newly identified therapeutic targets will be evaluated in adapted animal models. Eligible candidates should have experience with immunological and molecular biological and 'omics' techniques, and a strong motivation and team spirit. Medical students interested in an MD-PhD with a motivation for fundamental research are also suitable. German language skills would be helpful, solid English skills, written and spoken, are mandatory. The successful candidate should hold an above average Master degree or equivalent in biological sciences or related fields and should have a background in vascular biology.

The successful candidate will be working at the Departments of Angiology and Vascular Surgery at the Inselspital Bern with a joint Clinical Investigation Unit (CIU) and in the Department of Biomedical Research (DBMR) as a member of the research group 'Translational Angiology' under the direction of Prof. Dr. Yvonne Döring and Dr. S. Bernhard.
